The p.G885R variant (also known as c.2653G>A), located in coding exon 21 of the NF1 gene, results from a G to A substitution at nucleotide position 2653. The glycine at codon 885 is replaced by arginine, an amino acid with dissimilar properties. This alteration was reported as a variant of unknown signifiance in an individual with a sarcoma diagnosed at age 60 from a cohort of 1191 Asian individuals with cancer who underwent multi-gene panel testing (Chan GHJ et al. Oncotarget, 2018 Jul;9:30649-30660). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
