In summary, this is a novel in-frame deletion with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Experimental studies and prediction algorithms are not available for this variant, and the functional significance of the deleted amino acids is currently unknown. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a MLH1-related disease. This sequence change deletes 6 nucleotides from exon 12 of the MLH1 mRNA (c.1344_1349delGGGGGA). This leads to the deletion of 2 amino acid residues in the MLH1 protein (p.Glu448_Gly449del) but otherwise preserves the integrity of the reading frame. -

The c.1344_1349delGGGGGA variant (also known as p.E448_G449del) is located in coding exon 12 of the MLH1 gene. This variant results from an in-frame GGGGGA deletion at nucleotide positions 1344 to 1349. This results in the in-frame deletion of 2 residues (EG) including codons 448 and 449. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
