The c.263_267delAAGGA pathogenic mutation, located in coding exon 2 of the SMAD4 gene, results from a deletion of 5 nucleotides at nucleotide positions 263 to 267, causing a translational frameshift with a predicted alternate stop codon (p.K88Ifs*14). This alteration has been reported in families with juvenile polyposis syndrome (JPS) (Wain KE et al. Genet Med, 2014 Aug;16:588-93). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
Juvenile polyposis syndrome Pathogenic:1
This sequence change creates a premature translational stop signal (p.Lys88Ilefs*14) in the SMAD4 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SMAD4 are known to be pathogenic (PMID: 16152648, 16436638, 22810475). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with juvenile polyposis (PMID: 24525918). ClinVar contains an entry for this variant (Variation ID: 405496).
