Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. The p.Asp73Gly variant in POLE has not been previously reported in individuals w ith colorectal cancer but has been reported by other clinical laboratories in Cl inVar (Variation ID: 405626). It has also been identified in 1/111706 European c hromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstit ute.org). Computational prediction tools and conservation analysis suggest that the p.Asp73Gly variant may impact the protein, though this information is not pr edictive enough to determine pathogenicity.
