The c.1467_1468delCG variant, located in coding exon 14 of the POLE gene, results from a deletion of two nucleotides at nucleotide positions 1467 to 1468, causing a translational frameshift with a predicted alternate stop codon (p.D490Rfs*12). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. Loss-of-function variants subject to nonsense mediated decay (NMD) in POLE are known to cause POLE deficiency; however, such associations with POLE-related polymerase proofreading-associated polyposis (PPAP) have not been reported. Based on the supporting evidence, this alteration is pathogenic/likely pathogenic for POLE deficiency; however, the association of this alteration with POLE-related polymerase proofreading-associated polyposis (PPAP) is unknown. -
