This sequence change replaces proline with arginine at codon 1521 of the TSC2 protein (p.Pro1521Arg). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and arginine. This variant has been reported in an individual in the Leiden Open-source Variation Database (PMID: 21520333). However, in that individual a pathogenic allele was also identified in TSC2, which suggests that this c.4562C>G variant was not the primary cause of disease. In summary, this variant is a rare missense change with uncertain impact on protein function. While it is absent from the population and reported in an affected individual with a pathogenic variant, the available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
