Genes affected
SCN5A (HGNC:10593): (sodium voltage-gated channel alpha subunit 5) The protein encoded by this gene is an integral membrane protein and tetrodotoxin-resistant voltage-gated sodium channel subunit. This protein is found primarily in cardiac muscle and is responsible for the initial upstroke of the action potential in an electrocardiogram. Defects in this gene have been associated with long QT syndrome type 3 (LQT3), atrial fibrillation, cardiomyopathy, and Brugada syndrome 1, all autosomal dominant cardiac diseases. Alternative splicing results in several transcript variants encoding different isoforms. [provided by RefSeq, May 2022]

Likely pathogenic, criteria provided, single submitterclinical testingGenesis Genoma Lab, Genesis Genoma LabApr 26, 2022The c.2787+1G>T variant affects the donor splice site in intron 16 of the SCN5A gene and it is expected to disrupt mRNA splicing. It was identified in a patient with Brugada syndrome. -
not provided Pathogenic:1
Likely pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpDec 15, 2016In summary, donor and acceptor splice site variants are typically truncating (PMID: 16199547), and truncating variants in SCN5A are known to be pathogenic (PMID: 20129283, 22789973). However, without additional functional and/or genetic data, this variant has been classified as Likely Pathogenic. Algorithms developed to predict the effect of sequence changes on mRNA splicing suggest that this variant may alter mRNA splicing, but this prediction has not been confirmed by published transcriptional studies. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a SCN5A-related disease. This sequence change affects a donor splice site in intron 16 of the SCN5A gene. It is expected to disrupt mRNA splicing and likely results in an absent or disrupted protein product. -
