Algorithms developed to predict the effect of nucleotide changes on mRNA splicing suggest that this intronic variant may alter mRNA splicing, but this prediction has not been confirmed by published transcriptional studies. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a SMARCE1-related disease. This sequence change falls in intron 6 of the SMARCE1 mRNA. It does not directly change the encoded amino acid sequence of the SMARCE1 protein. In summary, this is a novel intronic change with uncertain impact on splicing. It has been classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Uncertain:1
The c.370-6T>C intronic variant results from a T to C substitution 6 nucleotides upstream from coding exon 6 in the SMARCE1 gene. This variant has been observed in at least one individual with a personal and/or family history that is consistent with SMARCE1-related meningioma susceptibility (Ambry internal data). In silico splice site analysis predicts that this alteration may weaken the native splice acceptor site; however, direct evidence is insufficient at this time (Ambry internal data). This nucleotide position is well conserved in available vertebrate species. Based on the available evidence, the clinical significance of this alteration remains unclear. -
