PVS1, PM2_supporting, PM5_supporting c.6272G>A, located in exon 43 of the ATM gene, is a nonsense variant expected to result in loss of function by premature protein truncation and nonsense-mediated mRNA decay (PVS1). It is not present in the population database gnomAD v2.1.1, non-cancer dataset (PM2_supporting). To our knowledge, neither relevant clinical data nor well-established functional studies have been reported for this variant. This variant has been reported in the ClinVar database (3x pathogenic) and has not been reported in LOVD. The premature termination codon is upstream of p.Arg3047* which is the most C-terminal pathogenic variant (PM5_Supporting). Based on currently available information, the variant cc.6272G>A should be considered a pathogenic variant according to ACMG Classification Rules Specified for ATM v1.1. -
The p.W2091* pathogenic mutation (also known as c.6272G>A), located in coding exon 42 of the ATM gene, results from a G to A substitution at nucleotide position 6272. This changes the amino acid from a tryptophan to a stop codon within coding exon 42. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
