Hereditary cancer-predisposing syndrome Uncertain:1Benign:1
Missense variant in a coldspot region where missense variants are very unlikely to be pathogenic (PMID:31911673). -
The p.E1036K variant (also known as c.3106G>A), located in coding exon 10 of the BRCA2 gene, results from a G to A substitution at nucleotide position 3106. The glutamic acid at codon 1036 is replaced by lysine, an amino acid with similar properties. This variant was detected in 1/251 German high risk breast and/or ovarian cancer patients; this patient also had a pathogenic mutation in the BRCA1 gene (Meyer P et al. Hum. Mutat. 2003 Sep; 22(3):259). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
not provided Uncertain:1
Observed in an individual with a personal history of breast and/or ovarian cancer and cervical cancer, who also harbored a pathogenic variant in BRCA1 (Meyer et al., 2003); In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; Not observed at significant frequency in large population cohorts (gnomAD); Also known as 3334G>A; This variant is associated with the following publications: (PMID: 29884841, 32377563, 12938098, 31911673) -
Hereditary breast ovarian cancer syndrome Uncertain:1
This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 1036 of the BRCA2 protein (p.Glu1036Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with breast and/or ovarian cancer (PMID: 12938098). This variant is also known as c.3334G>A. ClinVar contains an entry for this variant (Variation ID: 481544). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
