The p.E2082D variant (also known as c.6246A>C), located in coding exon 10 of the BRCA2 gene, results from an A to C substitution at nucleotide position 6246. The glutamic acid at codon 2082 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 2082 of the BRCA2 protein (p.Glu2082Asp). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 409573).
