Familial cancer of breast Pathogenic:2
This variant is considered pathogenic. This variant creates a frameshift predicted to result in premature protein truncation. -
This premature translational stop signal has been observed in individual(s) with breast cancer (PMID: 32658311). This sequence change creates a premature translational stop signal (p.Lys569Argfs*29) in the PALB2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PALB2 are known to be pathogenic (PMID: 17200668, 17200671, 17200672, 24136930, 25099575). This variant is not present in population databases (gnomAD no frequency). ClinVar contains an entry for this variant (Variation ID: 460908). For these reasons, this variant has been classified as Pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.1704_1707delAAAA pathogenic mutation, located in coding exon 5 of the PALB2 gene, results from a deletion of 4 nucleotides at nucleotide positions 1704 to 1707, causing a translational frameshift with a predicted alternate stop codon (p.K569Rfs*29). This alteration was seen in 2/732 breast cancer patients, 0/189 colorectal cancer patients and 0/490 cancer-free elderly controls in a Turkish population (Akcay IM et al. Int J Cancer, 2021 01;148:285-295). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
