The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in GALNT12 cause disease. Therefore, this variant has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a GALNT12-related disease. This sequence change results in a premature translational stop signal in the last exon of the GALNT12 mRNA at codon 547 (p.Cys547*). While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated GALNT12 protein. -
