The c.2317_2318delACinsCA variant, located in coding exon 16 of the TSC1 gene, results from an in-frame deletion of AC and insertion of CA at nucleotide positions 2317 to 2318. This results in the substitution of the threonine residue for a histidine residue at codon 773, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
