The serine at codon 2185 is replaced by glycine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. This alteration was reported in a cohort of Asian patients with multiple primary cancer diagnoses undergoing multigene panel testing for hereditary cancer susceptibility (Chan GHJ et al. Oncotarget. 2018 Jul;9:30649-30660). In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
