Mutations in the APC gene have been found to occur in most colorectal cancers, where disease-associated mutations tend to be clustered in a small region designated the mutation cluster region (MCR) and result in a truncated protein product. This changes the amino acid from a tryptophan to a stop codon within coding exon 4. This pathogenic mutation has been identified in two unrelated French patients with FAP (Lagarde A et al. J. Med. Genet. 2010 Oct;47:721-2; Olschwang S et al. Am. J. Hum. Genet. 1993 Feb;52:273-9). This pathogenic mutation has also been identified in a 56 year old female diagnosed FAP associated colon cancer who developed metastatic ovarian cancer eight years later (Crobach S et al. Fam. Cancer. 2012 Dec;11:671-3). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
