Variant summary
Our verdict is Pathogenic. The variant received 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate
The NM_177438.3(DICER1):c.2507_2508delTC(p.Phe836TyrfsTer8) variant causes a frameshift change involving the alteration of a conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Pathogenic (★). Synonymous variant affecting the same amino acid position (i.e. F836F) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

DICER1-related tumor predisposition Pathogenic:1
This sequence change deletes 2 nucleotides from exon 16 of the DICER1 mRNA (c.2507_2508delTC), causing a frameshift at codon 836. This creates a premature translational stop signal (p.Phe836Tyrfs*8) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in DICER1 are known to be pathogenic (PMID: 19556464, 21266384). For these reasons, this variant has been classified as Pathogenic. -
