ATM (HGNC:795): (ATM serine/threonine kinase) The protein encoded by this gene belongs to the PI3/PI4-kinase family. This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. This deletion of four nucleotides is denoted ATM c.4632_4635delCTTA at the cDNA level and p.Tyr1544Ter (Y1544X) at the protein level. The normal sequence, with the bases that are deleted in brackets, is AATA[delCTTA]GTGA. The deletion creates a nonsense variant, which changes a Tyrosine to a premature stop codon. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. ATM c.4632_4635delCTTA, previously reported as ATM 4630del4, has been observed in at least three Ataxia-Telangiectasia families, one of which included an observation of this variant in the homozygous state in an individual with Ataxia-Telangiectasia (Sandoval 1999, Laake 2000). This variant is considered likely pathogenic. -

The c.4632_4635delCTTA pathogenic mutation, located in coding exon 30 of the ATM gene, results from a deletion of 4 nucleotides at nucleotide positions 4632 to 4635, causing a translational frameshift with a predicted alternate stop codon (p.Y1544*). This alteration has been reported in multiple individuals with a clinical diagnosis of ataxia telangiectasia (Sandoval N et al. Hum. Mol. Genet. 1999 Jan;8(1):69-79; Laake K et al. Hum. Mutat. 2000 Sep;16(3):232-46). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
