The c.10183delG variant, located in coding exon 26 of the BRCA2 gene, results from a deletion of one nucleotide at nucleotide position 10183, causing a translational frameshift with a predicted alternate stop codon (p.E3395Rfs*32). This alteration occurs at the 3' terminus of the BRCA2 gene, is not expected to trigger nonsense-mediated mRNA decay and results in the elongation of the protein by 7 amino acids. This frameshift impacts the last 24 amino acids of the native protein. The exact functional effect of the altered amino acids is unknown. Based on the available evidence, the clinical significance of this variant remains unclear. -
