This in-frame deletion of three nucleotides in POLE is denoted c.2343_2345delGGC at the cDNA level and p.Ala782del (A782del) at the protein level. The normal sequence, with the bases that are deleted in brackets, is CGGC[delGGC]CGTG. This variant has not, to our knowledge, been published in the literature as a pathogenic or benign germline variant. POLE Ala782del was not observed in large population cohorts (Lek 2016). The deleted residue is located in the polymerase domain (Preston 2010). In silico analysis, which includes protein predictors and evolutionary conservation, supports a deleterious effect. Since in-frame deletions may or may not inhibit proper protein functioning, the clinical significance of this finding remains unclear at this time and we consider POLE Ala782del to be a variant of uncertain significance. -
This variant, c.2343_2345del, results in the deletion of 1 amino acid(s) of the POLE protein (p.Ala782del), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLE-related conditions. The c.2343_2345delGGC variant (also known as p.A782del) is located in coding exon 21 of the POLE gene. This variant results from an in-frame GGC deletion at nucleotide positions 2343 to 2345. This results in the in-frame deletion of an alanine at codon 782. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
