Genes affected
MYO7A (HGNC:7606): (myosin VIIA) This gene is a member of the myosin gene family. Myosins are mechanochemical proteins characterized by the presence of a motor domain, an actin-binding domain, a neck domain that interacts with other proteins, and a tail domain that serves as an anchor. This gene encodes an unconventional myosin with a very short tail. Defects in this gene are associated with the mouse shaker-1 phenotype and the human Usher syndrome 1B which are characterized by deafness, reduced vestibular function, and (in human) retinal degeneration. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2008]

Pathogenic, criteria provided, single submitterclinical testingWomen's Health and Genetics/Laboratory Corporation of America, LabCorpDec 18, 2023Variant summary: MYO7A c.1556G>A (p.Gly519Asp) results in a non-conservative amino acid change located in the Myosin head, motor domain (IPR001609) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. In addition, this variant disrupts the second nucleotide of exon 14, and therefore can affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.2e-05 in 248902 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1556G>A has been reported in the literature in both compound heterozygous and homozygous individuals affected with Usher Syndrome (e.g., Bharadwaj_2000, Roux_2006, Bonnet_2011, Zein_2014, Bujakowska_2014, Mansard_2021). These data indicate that the variant is very likely to be associated with disease. The following publications have been ascertained in the context of this evaluation (PMID: 10930322, 16679490, 21569298, 25425308, 25468891, 34948090). Five submitters have reported clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as pathogenic/likely pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpDec 14, 2023This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 519 of the MYO7A protein (p.Gly519Asp). This variant is present in population databases (rs111033206, gnomAD 0.006%). This missense change has been observed in individual(s) with Usher syndrome (PMID: 10930322, 16679490, 21569298, 21873662, 25468891). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 43151). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. For these reasons, this variant has been classified as Pathogenic. -
Rare genetic deafness Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineJun 23, 2016The p.Gly519Asp variant in MYO7A has now been identified in the homozygous or co mpound heterozygous state in at least 6 individuals with clinical features of Us her syndrome type I (Bharadwaj 2000, Roux 2006, Bonnet 2011, LMM data). It has b een identified in 2/66462 European chromosomes by the Exome Aggregation Consorti um (ExAC, http://exac.broadinstitute.org; dbSNP rs111033206). Although this vari ant has been identified in the general population, its frequency is low enough t o be consistent with the carrier frequency. In summary, this variant meets crite ria to be classified as pathogenic for autosomal recessive Usher syndrome. -
