Variant summary: GALT c.524G>A (p.Gly175Asp) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251496 control chromosomes (gnomAD). c.524G>A has been observed in several individuals affected with Galactosemia (e.g., Gort_2006, Boutron_2012, Coelho_2014). These data indicate that the variant is very likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, finding that the variant results in an almost complete loss of enzymatic activity (Coelho_2014). The following publications have been ascertained in the context of this evaluation (PMID: 17486650, 17041746, 22944367, 23749220, 25614870, 25814382).
