Genes affected
GALT (HGNC:4135): (galactose-1-phosphate uridylyltransferase) Galactose-1-phosphate uridyl transferase (GALT) catalyzes the second step of the Leloir pathway of galactose metabolism, namely the conversion of UDP-glucose + galactose-1-phosphate to glucose-1-phosphate + UDP-galactose. The absence of this enzyme results in classic galactosemia in humans and can be fatal in the newborn period if lactose is not removed from the diet. The pathophysiology of galactosemia has not been clearly defined. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2012]

Variant summary: GALT c.524G>A (p.Gly175Asp) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251496 control chromosomes (gnomAD). c.524G>A has been observed in several individuals affected with Galactosemia (e.g., Gort_2006, Boutron_2012, Coelho_2014). These data indicate that the variant is very likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, finding that the variant results in an almost complete loss of enzymatic activity (Coelho_2014). The following publications have been ascertained in the context of this evaluation (PMID: 17486650, 17041746, 22944367, 23749220, 25614870, 25814382). ClinVar contains an entry for this variant (Variation ID: 25203). Based on the evidence outlined above, the variant was classified as pathogenic. -
