The p.L376R variant (also known as c.1127T>G), located in coding exon 7 of the MEN1 gene, results from a T to G substitution at nucleotide position 1127. The leucine at codon 376 is replaced by arginine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6498 samples (12996 alleles) with coverage at this position. An alteration at this same amino acid position (p.L376P) was reported in a study of 200 unrelated Swedish index cases in a patient diagnosed with primary hyperparathyroidism, endocrine enterpancreatic tumor, and an anterior pituitary tumor. (Tham et al. J of Clinical Endocrinology & Metab. 2007. 92(9): 3389-3395). This amino acid position is completely conserved on sequence alignment. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. -
