In silico splice prediction models predict that c.-31+5 G>T damages the natural splice donor site for intron 1; however, the adjacent exon 1 is non-coding. Functional studies have shown that the c.-31+5 G>T variant reduces transcriptional activity and the expression of the BTK protein (Shin et al., 2008). In summary, we consider this variant to be likely pathogenic. -
