This changes the amino acid from a glutamic acid (E) to a stop codon at amino acid position 8802. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This exon is located in the A-band region of the N2-B isoform of the titin protein and is constitutively expressed in TTN transcripts (percent spliced in or PSI 100%). Based on data from gnomAD, the T allele has an overall frequency of <0.001% (1/216096) total alleles studied. The highest observed frequency was 0.001% (1/101728) of European (non-Finnish) alleles.
