Charcot-Marie-Tooth disease type 2 Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jan 19, 2023 | This missense change has been observed in individual(s) with hereditary motor neuropathy (PMID: 26392352). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt LMNA protein function. ClinVar contains an entry for this variant (Variation ID: 586129). This variant is present in population databases (no rsID available, gnomAD 0.006%). This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 318 of the LMNA protein (p.Ala318Val). - |

This variant has been reported in an individual affected with hypertrophic cardiomyopathy (Lopes 2015, thesis). This variant has also been identified in 2/248732 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Cardiovascular phenotype Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Oct 16, 2023 | The p.A318V variant (also known as c.953C>T), located in coding exon 6 of the LMNA gene, results from a C to T substitution at nucleotide position 953. The alanine at codon 318 is replaced by valine, an amino acid with similar properties. This alteration has been reported in a hypertrophic cardiomyopathy (HCM) cohort; however, clinical details were limited (Lopes LR et al. Heart, 2015 Feb;101:294-301). Additionally, this alteration was detected in an inherited peripheral neuropathy cohort and a muscular dystrophy cohort (Antoniadi T et al. BMC Med Genet, 2015 Sep;16:84; Masri AT et al. Clin Neurol Neurosurg, 2022 Jun;217:107271). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
