DNMT3B (HGNC:2979): (DNA methyltransferase 3 beta) CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. This gene encodes a DNA methyltransferase which is thought to function in de novo methylation, rather than maintenance methylation. The protein localizes primarily to the nucleus and its expression is developmentally regulated. Mutations in this gene cause the immunodeficiency-centromeric instability-facial anomalies (ICF) syndrome. Eight alternatively spliced transcript variants have been described. The full length sequences of variants 4 and 5 have not been determined. [provided by RefSeq, May 2011]

Variant summary: DNMT3B c.2177T>G (p.Val726Gly) results in a non-conservative amino acid change located in the C-5 cytosine-specific DNA methylase (Dnmt) domain (IPR001525) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251496 control chromosomes. c.2177T>G has been reported in the literature in biallelic individuals affected with ICF Syndrome, Type 1 (e.g. Weemaes_2013, Hansen_1999, Xu_1999). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in reduced methylation activity (e.g. Xie_2006, Ueda_2006). The following publications have been ascertained in the context of this evaluation (PMID: 10588719, 16501171, 23486536, 16543361, 10647011).
