This sequence change replaces threonine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 64 of the CAV3 protein (p.Thr64Asn). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CAV3-related conditions. This variant disrupts the p.Thr64 amino acid residue in CAV3. Other variant(s) that disrupt this residue have been observed in individuals with CAV3-related conditions (PMID: 11532985, 25214167), which suggests that this may be a clinically significant amino acid residue. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
