Experimental studies have shown that this missense change affects SOD1 function (PMID: 20184893, 20404910). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SOD1 protein function. ClinVar contains an entry for this variant (Variation ID: 14757). This missense change has been observed in individual(s) with autosomal dominant amyotrophic lateral sclerosis (PMID: 7655471, 8351519, 8446170, 9029070, 22647583). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 107 of the SOD1 protein (p.Leu107Val). -

This variant, also referred to as p.Leu106Val using legacy nomenclature, has previously report been reported in many unrelated families with ALS (Cudkowicz et al. 1997. PubMed ID: 9029070; Hineno et al. 2012. PubMed ID: 22647583; Kawamata et al. 1994. PubMed ID: 7911198). This variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. This variant is interpreted as pathogenic. -
