This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 2336 of the TG protein (p.Arg2336Gln). This variant is present in population databases (rs121912650, gnomAD 0.0009%). This missense change has been observed in individual(s) with TG-related conditions (PMID: 16720658, 29275168). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as R2317Q. ClinVar contains an entry for this variant (Variation ID: 12705). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TG protein function with a positive predictive value of 80%.
