Genes affected
PDGFRA (HGNC:8803): (platelet derived growth factor receptor alpha) This gene encodes a cell surface tyrosine kinase receptor for members of the platelet-derived growth factor family. These growth factors are mitogens for cells of mesenchymal origin. The identity of the growth factor bound to a receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, composed of both platelet-derived growth factor receptor alpha and beta polypeptides. Studies suggest that this gene plays a role in organ development, wound healing, and tumor progression. Mutations in this gene have been associated with idiopathic hypereosinophilic syndrome, somatic and familial gastrointestinal stromal tumors, and a variety of other cancers. ACMG classification

Classification made for transcript

Verdict is Likely_pathogenic. Variant got 7 ACMG points.

PM1
In a hotspot region, there are 2 aminoacids with missense pathogenic changes in the window of +-8 aminoacids around while only 1 benign, 8 uncertain in NM_006206.6
PM5
Other missense variant is known to change same aminoacid residue: Variant chr4-54285925-G-T is described in ClinVar as [Likely_pathogenic]. Clinvar id is 376250.Status of the report is no_assertion_criteria_provided, 0 stars.
PP2
Missense variant where missense usually causes diseases, PDGFRA
PP3
No computational evidence supports a deleterious effect, but strongly conserved according to phyloP
PP5
Variant 4-54285925-GA-AT is Pathogenic according to our data. Variant chr4-54285925-GA-AT is described in ClinVar as [Likely_pathogenic]. Clinvar id is 376055.Status of the report is no_assertion_criteria_provided, 0 stars.
