Predicted Consequence/Location: The variant is located in a mutational hot spot and/or well-established functional domain in which established pathogenic variants have been reported (PMID: 26637981, 24459181, 27631024). Missense variant. Missense changes are a common disease-causing mechanism. Damaging effect on gene or gene product predicted by in silico programs is uncertain [REVEL: 0.51 (damaging >=0.6, benign <0.4), 3Cnet: 0.62 (>0.75, sensitivity 0.96 and precision 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported as pathogenic/likely pathogenic with strong evidence (ClinVar ID: VCV000376247 /PMID: 22729224 /3billion dataset).
