Genes affected
PROC (HGNC:9451): (protein C, inactivator of coagulation factors Va and VIIIa) This gene encodes a vitamin K-dependent plasma glycoprotein. The encoded protein is cleaved to its activated form by the thrombin-thrombomodulin complex. This activated form contains a serine protease domain and functions in degradation of the activated forms of coagulation factors V and VIII. Mutations in this gene have been associated with thrombophilia due to protein C deficiency, neonatal purpura fulminans, and recurrent venous thrombosis.[provided by RefSeq, Dec 2009]

Thrombophilia due to protein C deficiency, autosomal dominant Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingInvitaeSep 23, 2022In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. ClinVar contains an entry for this variant (Variation ID: 948077). This variant is also known as 6265C>T (Q184Q). This variant has been observed in individual(s) with protein C deficiency (PMID: 7482420). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change affects codon 226 of the PROC mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the PROC protein. This variant also falls at the last nucleotide of exon 7, which is part of the consensus splice site for this exon. -
