The encoded protein is primarily composed of 22 spectrin repeats which are involved in dimer formation. It forms a component of the erythrocyte plasma membrane. This variant is present in population databases (rs121918636, gnomAD 0.007%). This missense change has been observed in individual(s) with SPTA1-related disorders (PMID: 2794061, 29729090). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is also known as p.Ser255Pro. ClinVar contains an entry for this variant (Variation ID: 12848).
