This sequence change replaces histidine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 418 of the CFI protein (p.His418Leu). This missense change has been observed in individual(s) with clinical features of CFI-related conditions (PMID: 8613545, 19065647, 32510551). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Studies have shown that this missense change alters CFI gene expression (PMID: 19065647, 32510551).
