Alternative splicing results in two transcript variants encoding different isoforms. Other alternatively spliced transcripts have been described, but the full length sequences have not been determined. Several of these transcripts are predicted to encode truncated products which lack the TM and may be secreted. In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as IVS46+653A>G. ClinVar contains an entry for this variant (Variation ID: 551996). Studies have shown that this variant results in the inclusion of a pseudoexon and introduces a premature termination codon (PMID: 19021639). This has been confirmed via cDNA sequencing of patient tissues, which showed the variant transcript had inclusion of a pseudoexon that results in a predicted frameshift: p.Gly2451fs (Michel-Calemard_2009). The variant was absent in 31404 control chromosomes (gnomAD). c.7350+653A>G has been reported in the literature in the compound heterozygous state in multiple individuals affected with Polycystic Kidney And Hepatic Disease (Michel-Calemard_2009). These data indicate that the variant is very likely to be associated with disease. The following publication has been ascertained in the context of this evaluation (PMID: 19021639). Four ClinVar submitters have assessed the variant since 2014: one classified the variant as likely pathogenic and three as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
