The c.8005_8007delAGA variant (also known as p.R2669del) is located in coding exon 17 of the BRCA2 gene. This variant results from an in-frame AGA deletion at nucleotide positions 8005 to 8007. This results in the in-frame deletion of an arginine at codon 2669. This nucleotide region is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
This variant causes an in-frame deletion of one amino acid of the BRCA2 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/250648 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
