Variant summary
Our verdict is Likely benign. The variant received -2 ACMG points: 0P and 2B. BP4BS2_Supporting
The NM_004360.5(CDH1):c.592G>A(p.Asp198Asn) variant causes a missense change. The variant allele was found at a frequency of 0.00000547 in 1,461,860 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. D198V) has been classified as Uncertain significance.

Hereditary cancer-predisposing syndrome Uncertain:2
The p.D198N variant (also known as c.592G>A), located in coding exon 5 of the CDH1 gene, results from a G to A substitution at nucleotide position 592. The aspartic acid at codon 198 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
