This missense change has been observed in individuals with X-linked myotubular myopathy (PMID: 9305655, 10215413, 10502779, 10790201, 11793470, 12522554, 18817572). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 182919 control chromosomes. c.721C>T has been reported in the literature in multiple individuals affected with Severe/Mild X-Linked Myotubular Myopathy (example, Laporte_2001, Laporte_2000, Bachman_2017). These data indicate that the variant is very likely to be associated with disease. Several publications report experimental evidence evaluating an impact on protein function, reporting disruped interaction between myotubularin and MTMR12 proteins (example, Gupta_2013). Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
