The encoded protein, which belongs to the tumor necrosis factor family, acts as a homotrimer and may be involved in cell-cell signaling during the development of ectodermal organs. Defects in this gene are a cause of ectodermal dysplasia, anhidrotic, which is also known as X-linked hypohidrotic ectodermal dysplasia. Proline (Pr o) at position 209 is highly conserved in mammals and evolutionarily distant spe cies, supporting that a change at this position may not be tolerated. Additional computational prediction tools suggest that this variant may impact the protein , though this information is not predictive enough to determine pathogenicity. I n summary, while there is some suspicion for a pathogenic role, the clinical sig nificance of the Pro209Leu variant is uncertain. -
