KRIT1-related disorder Pathogenic:1
The KRIT1 c.1201C>T variant is predicted to result in premature protein termination (p.Gln401*). This variant was reported in an individual with cerebral cavernous malformations and seizures (Feldmeyer et al. 2014. PubMed ID: 24721395). This variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Nonsense variants in KRIT1 are expected to be pathogenic. This variant is interpreted as pathogenic. -
Cerebral cavernous malformation Pathogenic:1
This sequence change creates a premature translational stop signal (p.Gln401*) in the KRIT1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in KRIT1 are known to be pathogenic (PMID: 10508515, 11222804, 12404106, 24689081). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with multiple cerebral cavernomas and hyper-keratotic angioma (PMID: 24721395). ClinVar contains an entry for this variant (Variation ID: 468205). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
