Genes affected
CDKN1C (HGNC:1786): (cyclin dependent kinase inhibitor 1C) This gene is imprinted, with preferential expression of the maternal allele. The encoded protein is a tight-binding, strong inhibitor of several G1 cyclin/Cdk complexes and a negative regulator of cell proliferation. Mutations in this gene are implicated in sporadic cancers and Beckwith-Wiedemann syndorome, suggesting that this gene is a tumor suppressor candidate. Three transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Oct 2010]

The CDKN1C c.78C>T variant is not predicted to result in an amino acid change (p.=). This variant is predicted to interfere with splicing. To our knowledge, this variant has not been reported in the literature. It was observed in 1 out of ~31390 alleles in gnomAD (https://gnomad.broadinstitute.org/variant/11-2906642-G-A). In ClinVar, this variant is interpreted as likely benign (https://preview.ncbi.nlm.nih.gov/clinvar/variation/524736/). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
