Mutations of this gene has been associated with autosomal recessive HEM/Greenberg skeletal dysplasia. This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 119 of the LBR protein (p.Pro119Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Pelger-Huet anomaly (PMID: 14617022). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 9530).
