The c.3791delC pathogenic variant in the TSC2 gene has been reported previously in association with TSC (TSC2 LOVD). The deletion causes a frameshift starting with codon Proline 1264, changes this amino acid to a Leucine residue and creates a premature Stop codon at position 61 of the new reading frame, denoted p.Pro1264LeufsX61. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.3791delC variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). -

The c.3791delC pathogenic mutation, located in coding exon 30 of the TSC2 gene, results from a deletion of one nucleotide at nucleotide position 3791, causing a translational frameshift with a predicted alternate stop codon (p.P1264Lfs*61). This variant was reported in an individual with features consistent with tuberous sclerosis complex (Togi S et al. Int J Mol Sci, 2022 Sep;23:). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
