This variant is reported in 0.012% of alleles in individuals of African descent in gnomAD. Variants that disrupt the consensus splice donor site in BBS9 are expected to be pathogenic. This variant is interpreted as pathogenic. -

Studies have shown that disruption of this splice site results in insertion of 4 nucleotides between exons 3 and 4 and introduces a new termination codon (PMID: 24849935). Disruption of this splice site has been observed in individual(s) with Bardet-Biedl syndrome (PMID: 24849935). This variant is present in population databases (rs137962929, gnomAD 0.01%). This sequence change affects a donor splice site in intron 3 of the BBS9 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely disrupts the C-terminus of the protein. -
