The C719R variant hof uncertain significance in the MYBPC3 gene as not been published as pathogenic or been reported as benign to our knowledge. This variant is observed in 3/17,096 alleles from individuals of East Asian ancestry in large population cohorts (Lek et al., 2016). The C719R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, in-silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function.Therefore, based on the currently available information, it is unclear whether this variant is pathogenic or benign. -
Hypertrophic cardiomyopathy Uncertain:1
This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 719 of the MYBPC3 protein (p.Cys719Arg). This variant is present in population databases (no rsID available, gnomAD 0.02%). This missense change has been observed in individual(s) with sudden death with suspected hypertrophic cardiomyopathy (PMID: 29219260). ClinVar contains an entry for this variant (Variation ID: 546308). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Studies have shown that this missense change does not affect mRNA splicing (PMID: 34097875).
