The variant allele was found at a frequency of 0.0007 in 251296 control chromosomes, predominantly at a frequency of 0.0012 within the Non-Finnish European subpopulation in the gnomAD database. The observed variant frequency within Non-Finnish European control individuals in the gnomAD database is approximately 4-fold of the estimated maximal expected allele frequency for a pathogenic variant in DOCK8 causing Severe Combined Immunodeficiency phenotype (0.00035), strongly suggesting that the variant is a benign polymorphism found primarily in populations of Non-Finnish European origin. c.3312G>C has been reported in the literature in individuals affected with Common Variable Immunodeficiency and Autism/Developmental Delay, without strong evidence for causality (Maffucci_2016, Guo_2018). These report(s) do not provide unequivocal conclusions about association of the variant with Severe Combined Immunodeficiency.
