This alteration results from an A to G substitution at nucleotide position 763, causing the lysine (K) at amino acid position 255 to be replaced by a glutamic acid (E). Based on data from the Genome Aggregation Database (gnomAD) database, the SPG11 c.763A>G alteration was observed in 0.01% (39/282828) of total alleles studied, with a frequency of 0.14% (34/24962) in the African subpopulation. The p.K255 amino acid is not conserved in available vertebrate species. The p.K255E alteration is predicted to be tolerated by in silico analysis.
