TECTA (HGNC:11720): (tectorin alpha) The tectorial membrane is an extracellular matrix of the inner ear that contacts the stereocilia bundles of specialized sensory hair cells. Sound induces movement of these hair cells relative to the tectorial membrane, deflects the stereocilia, and leads to fluctuations in hair-cell membrane potential, transducing sound into electrical signals. Alpha-tectorin is one of the major noncollagenous components of the tectorial membrane. Mutations in the TECTA gene have been shown to be responsible for autosomal dominant nonsyndromic hearing impairment and a recessive form of sensorineural pre-lingual non-syndromic deafness. Although this variant has been seen in the general population, its frequency is not high enoug h to rule out a pathogenic role. Computational prediction tools and conservation analyses suggest that the variant may not impact the protein, though this infor mation is not predictive enough to rule out pathogenicity. Of note, the leucine (Leu) at position 943 is conserved in mammals but several fish species carry a m ethionine (Met) at this position, raising the possibility this change may be tol erated. In summary, the clinical significance of the p.Leu943Met variant is unce rtain. -
