The p.L135Q variant (also known as c.404T>A), located in coding exon 3 of the BRCA2 gene, results from a T to A substitution at nucleotide position 404. The leucine at codon 135 is replaced by glutamine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Hereditary breast ovarian cancer syndrome Uncertain:1
This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. This sequence change replaces leucine with glutamine at codon 135 of the BRCA2 protein (p.Leu135Gln). The leucine residue is weakly conserved and there is a moderate physicochemical difference between leucine and glutamine. This variant is not present in population databases (ExAC no frequency).
